Biotherapeutic size variants can dramatically influence pharmacokinetics, efficacy, safety, and stability—making precise profiling of protein size a cornerstone of biopharmaceutical development and quality control. 

Chia Thach (Bio-Techne) will highlight the latest advancements in CE-SDS using the Maurice Platform, which has been replacing manual SDS-PAGE methods for biotherapeutic characterization due to its ease of use and robust workflow. He will discuss specific workflows for size and purity analysis of Antibody-Drug Conjugates (ADCs) and Adeno-Associated Virus (AAVs). He will share recent data demonstrating how throughput and productivity can be significantly improved using the Turbo CE-SDS cartridges.

Our featured speaker, Caroline Daniels (Shattuck Labs) will describe a workflow using the Maurice system to separate structurally similar molecules based on the number of N-linked glycosylation sites. After evaluating both the Maurice CE-SDS Plus and Turbo CE-SDS cartridges for routine analysis of highly glycosylated heterodimeric fusion proteins, her team selected the Turbo cartridge to reduce run time by 75% while maintaining resolution.

At Shattuck labs, this method is used to support process development decisions to enrich for the bispecific protein, as well as to support GMP release and stability. This strategy can be readily applied to other heterodimeric or bispecific glycoproteins, provided there are sufficient differences in glycosylation between the polypeptide chains.
